William was the son and heir of Henry Wollaston, a wealthy draper in London, and his first wife Sarah Burgis. He inherited extensive landholdings in Staffordshire including Oncote Manor, where he settled after his father's death in 1616. Oncote did not suit him, so he bought Shenton Manor in Leicestershire in 1626, which he rebuilt in Jacobean style in 1629. He was appointed High Sheriff of Leicestershire and Staffordshire, and purchased additional estates in Leicestershire, Derbyshire and Suffolk. He was considered to be a good landlord, and was thought to have had Royalist sympathies.
William married Ann Whitgreve (qv) in 1616, and had four sons. Their eldest son Henry was educated at Market Bosworth School, Trinity College Cambridge, and the Inner Temple; after his marriage to Elizabeth Keightley he lived in Finborough Hall in Suffolk, but did not have any children and did not survive his father. Their next son was William (qv) who was his father's heir. They also had twins (John and Richard) who died in infancy.
